Elizabeth a medical student at Lady Margaret Hall working with Professor Robert Maclaren's reseach team in a project developing and testing structure-guided chemical modifications of SaCas9 gRNA. Chemically modified SpCas9 gRNA has been shown to effect up to 80% CRISPR editing in vivo in mouse liver, but no data currently exists for chemical modification of SaCas9 gRNA. SaCas9 is a highly desirable system to optimise, due to its smaller size and thus greater ease of packaging within an adeno-associated viral (AAV) vector.
